About The Position

Working independently, under general supervision, performs cutting-edge research projects involving the use of CRISPR-Cas9 high-throughput screening to understand genetic disease and identify gene therapy strategies in the Sherwood lab (https://sherwoodlab.bwh.harvard.edu/). This person will be in charge of all aspects of the projects for which they are responsible, which will include: stem cell culture; precision CRISPR-Cas9 genome editing techniques; genomic analysis; molecular biology; data analysis; and writing scientific papers to communicate salient findings. A strong academic background in genetics and CRISPR-Cas9 is expected, and prior research experience at the undergraduate level or higher is required. Technicians are given autonomy to lead projects, and previously trained technicians have co-authored journal articles and moved on to MD and PhD programs. While the primary responsibilities are wet lab experimental, the lab also has a strong computational component, so computational background is helpful but not essential. Performs experimental laboratory work in the field of genomics, stem cell biology, and genome editing. Full project management requires integrated performance of all of these tasks on a daily basis along with organizational skills to balance project demands. Previously trained technicians have co-authored journal articles and moved on to MD and PhD programs, so we are looking for someone who could commit 2 years to an exciting research position to prepare them for graduate/medical school.

Requirements

  • B.S. in a biological/physical science required.
  • Lab experience is required, experience with mammalian cell culture, molecular cloning, and genomic work is preferred.
  • Must have a sound understanding of laboratory policies and procedures - usually acquired through one-three months on-the-job training.
  • Must have good manual dexterity.
  • Must demonstrate careful attention to detail.
  • Must be able to maintain a neat and orderly working environment.

Nice To Haves

  • computational background is helpful but not essential.

Responsibilities

  • Molecular biology including devising and carrying out plasmid cloning strategies.
  • Cell culture including routine maintenance, genetic manipulation, and high-throughput CRISPR-Cas9 screening.
  • Genome editing-based screens including library production, testing, genomic library preparation and data analysis.
  • Genomic procedures including ChIP-seq, DNase-seq, single cell RNA-seq, PCR, and library preparation.
  • Creative employment of novel laboratory techniques as the project demands.
  • Maintain basic records of methodologies and test results in a log of work performed
  • Identify unusual findings and report such findings to supervisor
  • Maintain equipment and supplies in a neat and orderly manner
  • Maintain established department policies, procedures, objectives, quality assurance, safety, environmental and infection control
  • Maintain established hospital policies, procedures, and objectives for laboratory safety
  • Performs other duties and responsibilities as directed.
